Rio Negro (bahasa Portugis: Rio Negro [ˈʁi.u nɛɡɾu]; bahasa Spanyol: Río Negro [ˈri.o ˈneɣɾo]) merupakan sungai berwarna hitam terbesar yang merupakan anak sungai dari Sungai Amazon. Sesuai dengan namanya Rio Negro yang berarti “Sungai Hitam,” meskipun tidak sehitam yang dibayangkan bahkan terlihat seperti teh kental. Warna hitam tersebut berasal dari asam organik yang tidak terurai dengan sempurna oleh vegetasi yang ada di sekitar sungai. Di Kolombia, Sungai Rio Negro pun dikenal dengan nama Sungai Guainía, sungainya mengalir melalui Rio Solimões menuju Sungai Amazon wilayah Manaus, Brasil.
